What version of picotool are you using? You need to be using picotool version 2.2.0 or greater, as that’s when the cyw43-firmware family ID was added.

That allowed some more steps, up to:
> picotool load picow_ota_update_wifi_firmware.uf2
Family ID 0xe48bff55 cannot be downloaded anywhere
ERROR: This file cannot be loaded into the partition table on the device

This error is because you changed the partition table JSON, so it would no longer accept the cyw43-firmware family ID of 0xe48bff55
